傅向东研究组发现G蛋白调控稻米品质和产量的全新分子机制

长期以来我国水稻育种的主要目的是提高产量,但是随着人们生活水平不断提高,对稻米品质也提出了更高的要求。然而,高产水稻品种的品质往往相对较差,而优质水稻的产量也往往相对较低。如何解决“高产不优质,优质不高产”的矛盾一直是水稻育种面临的难题。

 

中国科学院遗传与发育生物学研究所傅向东研究组在水稻优质和高产性状协同改良的研究中取得重要进展。从长粒型美国粳稻品种L204中成功分离并克隆了一个控制稻米产量和品质协同提升的重要基因LGY3,该基因编码一个MIKC型MADS-box家族蛋白OsMADS1。研究发现,OsMADS1转录因子可以直接与G蛋白γ亚基DEP1和GS3蛋白互作,共同调控下游靶基因的表达。目前人们对植物细胞G蛋白信号转导通路的认识还非常有限,该项研究揭示了G蛋白通过与MADS转录因子互作的全新分子调控机制,为深入研究G蛋白信号转导途径的分子基础提供了新的切入点。

 

通过水稻种质资源测序分析发现,在尼瓦拉野生稻和热带粳稻中存在一种新的变异类型,编码一个C端截短的蛋白OsMADS1lgy3。研究还表明,该等位变异可以让稻米变得更加细长,有效减少垩白率和垩白面积,显著提升了稻米在外观、口感等方面的品质。然而,在我国大面积种植的高产水稻品种中却不含这种自然变异类型。将等位基因OsMADS1lgy3引入我国现有高产杂交水稻后,在显著提升稻米品质的基础上还可使其产量增加7%以上;将该等位基因OsMADS1lgy3和高产基因dep1聚合并应用到常规稻育种中,不仅可显著提升稻米品质,还可进一步提高水稻产量10%以上。DEP1-GS3-OsMADS1分子模块的发现和应用有望解决水稻高产与优质之间的矛盾,未来可用于“超级稻”新品种培育,使得稻米能够同时好吃又高产。

 

该研究成果于2018年2月27日在线发表于Nature Communications (DOI:10.1038/s41467-018-03047-9)。本项目得到国家自然基金委和中科院分子模块设计育种创新体系先导科技专项资助。
