新型水凝胶可按病情释放药量

英国《自然·通讯》杂志近日发表了一项生物医学工程最新成果:美国科学家利用小鼠模型成功展示了一种可响应疾病活动性的释药凝胶,其可装载不同的小分子,能“聪明”地根据疾病发作的不同严重程度释放相应的剂量,或将开启未来的新型治疗方案。

 

很多疾病的发作情况是时轻时重的,而目前传递剂量保持不变的药物治疗方法,很可能无法达到最优用量,容易贻误病情或过度治疗。

 

有鉴于此,美国布莱根妇女医院研究人员杰弗里·卡普及其同事研发出一种响应发作的水凝胶,可装载不同的小分子,根据疾病发作的严重程度释放相应剂量的药物。他们在炎症性关节炎的细胞和小鼠模型中进行实验,当凝胶接触到关节炎相关酶时,便会分解。释放出一种用于治疗关节炎的皮质类固醇,这时可观察到关节炎活动性降低。

 

关节炎病因十分复杂,虽然并不致命,但严重者可导致关节残疾,影响患者生活质量。而且,炎症性关节炎患者的病情通常一阵子缓,一阵子急,急的时候也就是疾病发作。研究团队称,响应型水凝胶是一种潜在的治疗炎症性关节炎的有效方法。

 

但研究人员也谨慎提出,需要更多的研究来优化凝胶的成分,并有必要在有人类大小关节的大型动物上测试,之后才可以考虑人类临床试验。他们认为,这项成果或是医学界设计响应疾病活动性治疗方案迈出的第一步。
