植物所科研人员揭示水稻耐寒调节新途径

       全球气候变化引起的局部温度异常直接威胁作物生产。针对作物耐受低温的机制进行研究,有利于基于分子设计的作物遗传改良工作的开展。目前,水稻耐寒信号转导途径框架业已建立,但其成员间的调节机制却知之不多。 

  中国科学院植物研究所种康院士团队针对OsbHLH002为核心的调控途径开展研究,揭示了调控水稻对低温响应和耐受的新途径。OsbHLH002是水稻bHLH转录因子家族一百多个成员之一,是耐寒信号途径中的核心成员。该蛋白与拟南芥ICE1蛋白同源性最高,因此也被称作OsICE1。研究人员近期的工作证明,有丝分裂原蛋白激酶OsMAPK3和泛素连接酶OsHOS1能够精细调节OsbHLH002的活性与积累,进而导致渗透保护剂海藻糖的合成,从而赋予水稻耐寒性。 

  研究发现,当水稻遇到低温胁迫时,OsMAPK3被激活,通过直接磷酸化OsbHLH002增强后者的转录激活能力。同时,此二者之间的相互作用抑制了OsHOS1与OsbHLH002之间的相互作用,进而减少了OsbHLH002的泛素化和降解过程。不断累积的高活性OsbHLH002有效激活了海藻糖-6-磷酸磷酸酶基因OsTPP1的表达,水解海藻糖-6-磷酸,提高渗透保护剂海藻糖含量,增强水稻的耐寒性。 

  该研究通过转录因子OsbHLH002,建立起激酶级联信号、渗透保护物质和非生物胁迫之间的联系,揭示了OsMAPK3—OsbHLH002—OsTPP1调控水稻对低温响应和耐受的新途径,对于水稻分子设计育种具有重要理论意义。 

  该成果18日发表于国际学术刊物Developmental Cell上。种康研究组博士后张泽勇为论文第一作者,徐云远研究员和种康研究员为共同通讯作者。相关工作得到了国家农业部、中国科学院和国家基金委的资助。
